The amount of enzyme required to degrade 1 μg plasmid DNA at 37 ℃ for 10 min was defined as one active unit.

Remove the genomic DNA from sample.
1.Prepare the following reaction mixture in a microcentrifuge tube.
2.Incubate for 20 - 30 min at 37 ℃.
3.Dissolve the precipitate in a suitable amount of DEPC-treated water. Confirm the genomic DNA is removed completely by agarose electrophoresis and measure the RNA concentration. When the genomic DNA is not removed completely, increase the amount of enzyme, or extend reaction time.
Compare the digestion activity of different dilution of DNase I from D2P or other vendor. Both of them can digest 1 μg pUC19 plasmid completely in 37 ℃ for 5 min.
